Asparaginase is an enzyme used in clinical treatments as a chemotherapeutic agent and in food technology to prevent acrylamide formation in fried and baked foods. Asparaginase is industrially produced by microorganisms, mainly gram-negative bacteria. Zymomonas mobilis is a Gram-negative bacterium that utilizes glucose, fructose and sucrose as carbon source and has been known for its efficiency in producing ethanol, sorbitol, levan, gluconic acid and has recently aroused interest for asparaginase production. Current assay optimizes the production of Z. mobilis asparaginase by continuous fermentation using response surface experimental design and methodology. The studied variables comprised sucrose, yeast extract and asparagine. Optimized condition obtained... |

Protein hydrolysate was prepared from common cuttlefish Sepia officinalis by Pepsin. Hydrolysis conditions (time, temperature, and enzyme activity) were optimized by response surface methodology (RSM) using a factorial design. The regression coefficient was close to 0.999, observed during both experimental and validation runs, and indicated the validity of prediction model. An enzyme activity to substrate ratio (X1), 0.04 AU/g protein; time (X2), 85 minutes; and temperature (X3), 45 °C were found to be the optimum conditions for a higher degree of hydrolysis (21%) and nitrogen recovery (90%) using Pepsin. According to amino acid analysis results and chemical score, methionine and histidine are the limiting amino acids in the hydrolysates, in that order.... |

The production of tannase by Aspergillus sp. GM4 under solid-state fermentation (SSF) was investigated using different vegetables leaves such as mango, jamun, coffee and agricultural residues such as coffee husks, rice husks and wheat bran. Among substrates used jamun leaves yielded high tannase production. The Plackett-Burman design was conducted to evaluate the effects of 12 independent variables on the production of tannase under SSF using jamun leaves as substrate. Among these variables, incubation time, potassium nitrate and tannic acid had significant effects on enzyme production. The best incubation time was studied and others variables were optimized using the Central Composite Design. The best conditions for tannase production were: incubation... |

It was intended with the present work to verify the effect of phytase on the performance, bone mineralization and content of plasma urea of swine. 80 swine (30 ± 3.1 kg/LW), crossbred (LD x LW) males and females, allocated to a randomized block design (RBD) with four treatments and five replicates. The treatments consisted of a growing swine diet on the basis of corn, soybean meal and defatted rice bran (DRB) supplemented with four levels of phytase (0, 400, 800 and 1200 FTU/kg) The diet was formulated to meet the requirements of growing swine except for available calcium and phosphorus, the levels of which were 0.64 and 0.25%, respectively. The enzyme phytase utilized was Natuphos 5000. Over the experimental period every eight days, blood of two animals... |

ABSTRACT: In order to evaluate the efficiency of phytase in diets with low and high phytate phosphorus (PP) 0content, as a consequence of wheat bran inclusion, on the relative weight of organs, intestinal morphometry and performance, three hundred and eighty-four male Cobb500 broilers were housed in metabolic cages. Animals were assigned into four treatments in a 2x2 factorial scheme in a randomized block design with eight replicates of 12 birds each. From 11 days of age birds received experimental diets, which consisted of: Diet low in PP; Diet low in PP with phytase (500FTU kg-1); Diet with a high PP and Diet with a high PP with phytase (500FTU kg-1). At 22 and 32 days of age two birds were slaughtered in order to collect gizzard, heart, liver, cecum,... |

Them aim scope of this study is to characterize the enzymatic polymorphism found in the Melipona quadrifasciata Lepeletier, 1936 populations from Ribeirão Preto, São Paulo and Espírito Santo, Brazil and its hybrids. Samples from each colony (about 52) were prepared for starch gel electrophoresis in order to investigate the genetic variation of the following enzimes: esterase (EST), isocitrate dehydrogenase (IDH), malic enzyme (ME), phosphoglucomutase (PGM), superoxide desmutase (SOD), α-glycerophosphate dehydrogenase (αPGD), malate dehydrogenase (MDH), leucine aminopeptidase (LAP), hexokinase (HK) and phosphoglucose isomerase (PGI). The analysis showed that LAP and HK did not show enzymatic activity and EST showed two alleles(est-sand and est-f) while all... |

The strain Chryseobacterium sp. kr6 shown to be useful for biotechnological purposes such as hydrolysis of poultry feathers and de-hairing of bovine pelts. The effect of temperature, initial pH and media composition on protease production by this keratinolytic strain was studied. The enzyme was produced between 25 and 37ºC, with maximum activity and yield at 30ºC. When protease production was tested in media with different initial pH, maximum activity was observed when cultivation was carried out at 30ºC and initial pH ranging from 6.0 to 8.0. Higher activity was observed when feathers or feather meal were used as growth substrates, followed by soybean meal. The addition of carbohydrates or surfactants to feather broth resulted in decrease in keratinolytic... |

ABSTRACT The objective of the current study was to evaluate the effects of three calcium:available phosphorus (Ca:aP) ratios and different supplementation with phytases from different origins on performance and ash, Ca, and P contents in the tibia of broilers. A total of 900 male broilers (22 to 35 days old) were fed ten dietary treatments in a completely randomized design. A 3 × 3 + 1 factorial scheme was used, corresponding to three Ca:aP ratios (4.5:1.0, 6.0:1.0, and 7.5:1.0) and three different supplementations with phytases from different origins (isolated or combined supplementation with bacterial and fungal phytase), plus a control diet. Regardless of the dietary Ca:aP ratio, the isolated use of bacterial phytase provided better feed conversion than... |

Whole wheat flour (WWF) contains various kinds of functional ingredients, such as dietary fiber, resistant starch, minerals, etc., and bread containing WWF is in great demand in many countries because of its health benefits. Although dough with WWF often results in lower quality bread, previous studies suggested that this could be improved by the addition of suitable enzymes. This study investigated the effects of WWF substitution and enzyme treatments using alpha-amylase (AM) and hemicellulase (HC) on bread making qualities. Results showed that the addition of WWF produced dough with low gas retention of dough (GRD) and specific loaf volume (SLV). However, AM and HC drastically improved both GRD and SLV of WWF-substituted dough and bread by degrading... |
