Optimization of asparaginase production from Zymomonas mobilis by continuous fermentation Biological Sciences

Asparaginase is an enzyme used in clinical treatments as a chemotherapeutic agent and in food technology to prevent acrylamide formation in fried and baked foods. Asparaginase is industrially produced by microorganisms, mainly gram-negative bacteria. Zymomonas mobilis is a Gram-negative bacterium that utilizes glucose, fructose and sucrose as carbon source and has been known for its efficiency in producing ethanol, sorbitol, levan, gluconic acid and has recently aroused interest for asparaginase production. Optimization of common cuttlefish (Sepia officinalis) protein hydrolysate using Pepsin by Response Surface Methodology ArchiMer

Protein hydrolysate was prepared from common cuttlefish Sepia officinalis by Pepsin. Hydrolysis conditions (time, temperature, and enzyme activity) were optimized by response surface methodology (RSM) using a factorial design. The regression coefficient was close to 0.999, observed during both experimental and validation runs, and indicated the validity of prediction model. An enzyme activity to substrate ratio (X1), 0.04 AU/g protein; time (X2), 85 minutes; and temperature (X3), 45 °C were found to be the optimum conditions for a higher degree of hydrolysis (21%) and nitrogen recovery (90%) using Pepsin. The production of tannase by Aspergillus sp. GM4 under solid-state fermentation (SSF)  was investigated using different vegetables leaves such as mango, jamun, coffee and agricultural residues such as coffee husks, rice husks and wheat bran. Among substrates used jamun leaves yielded high tannase production. The Plackett-Burman design was conducted to evaluate the effects of 12 independent variables on the production of tannase under SSF using jamun leaves as substrate. It was intended with the present work to verify the effect of phytase on the performance, bone mineralization and content of plasma urea of swine. 80 swine (30 ± 3.1 kg/LW), crossbred (LD x LW) males and females, allocated to a randomized block design (RBD) with four treatments and five replicates. ABSTRACT: In order to evaluate the efficiency of phytase in diets with low and high phytate phosphorus (PP) 0content, as a consequence of wheat bran inclusion, on the relative weight of organs, intestinal morphometry and performance, three hundred and eighty-four male Cobb500 broilers were housed in metabolic cages. Animals were assigned into four treatments in a 2x2 factorial scheme in a randomized block design with eight replicates of 12 birds each. Production and characterization of galactose oxidase produced by four isolates of Fusarium graminearum BABT

A screening aimed to find new galactose oxidase producer isolates and to evaluate the production among Fusarium graminearum strains was conducted. Thirty-five isolates out of 39 analysed produced the enzyme at several levels. The data indicated a wide distribution of galactose oxidase within F. graminearum and also revealed new producer isolates. The enzyme produced by different isolates showed similar thermal activity and stability and were active on same substrates. However, the optimum pH ranged from 7.0 to 7.5. Thus, all evaluated isolates were suitable for the production of galactose oxidase.

Production of an extracellular keratinase from Chryseobacterium sp. growing on raw feathers Electron. J. Biotechnol.

The strain Chryseobacterium sp. kr6 shown to be useful for biotechnological purposes such as hydrolysis of poultry feathers and de-hairing of bovine pelts. The effect of temperature, initial pH and media composition on protease production by this keratinolytic strain was studied. The enzyme was produced between 25 and 37ºC, with maximum activity and yield at 30ºC. When protease production was tested in media with different initial pH, maximum activity was observed when cultivation was carried out at 30ºC and initial pH ranging from 6.0 to 8.0. Higher activity was observed when feathers or feather meal were used as growth substrates, followed by soybean meal. Production of tannase by Aspergillus tamarii in submerged cultures BABT

The production of tannase by Aspergillus tamarii was studied in submerged cultures. The fungus produced an extracellular tannase after two days of growth in mineral medium containing tannic acid, gallic acid and methyl gallate as carbon source. The best result was obtained using gallic acid as inducer (20.6 U/ml). The production of enzyme was strongly repressed by the presence of glucose. Crude enzyme was optimally active at pH 5.0 and 30º C. The enzyme was stable in a large range of pH and up to the temperature of 45º C.

Purification and characterisation of trypsin-like enzyme from the pyloric caeca of cod (Gadus morhua) II BABT

A trypsin -like enzyme from the pyloric caeca of cod (Gadus morhua) was purified by affinity chromatography on CHOM Sepharose 4B. Some characteristics were established by its catalytic activity on T.A.M.E., typical enzyme substrate, and serine protease inhibitors. The enzyme had an isoelectric point of 5.30 and 5.89 and was very similar in amino acid composition to bovine trypsin, but differed in having a higher relative amount of acidic amino acids and a lower amount of basic amino acids. The enzyme also hydrolysed fish protein substrates.

Eleusine indica (goosegrass) is a diploid grass weed which has developed resistance to ACCase inhibitors during the last ten years due to the intensive and frequent use of sethoxydim to control grass weeds in soybean crops in Brazil. Plant dose-response assays confirmed the resistant behaviour of one biotype obtaining high resistance factor values: 143 (fenoxaprop), 126 (haloxyfop), 84 (sethoxydim) to 58 (fluazifop). ACCase in vitro assays indicated a target site resistance as the main cause of reduced susceptibility to ACCase inhibitors. Whole wheat flour (WWF) contains various kinds of functional ingredients, such as dietary fiber, resistant starch, minerals, etc., and bread containing WWF is in great demand in many countries because of its health benefits. Although dough with WWF often results in lower quality bread, previous studies suggested that this could be improved by the addition of suitable enzymes. This study investigated the effects of WWF substitution and enzyme treatments using alpha-amylase (AM) and hemicellulase (HC) on bread making qualities. Use of vinasse and sugarcane bagasse for the production of enzymes by lignocellulolytic fungi BABT

In this present work, three strains of Pleurotus and Trichoderma reesei were cultivated in media with pre-treated bagasse and vinasse. Cellulolytic and lignolytic activities and biomass production were analyzed. The treatment of the bagasse with 2% H2O2 + 1.5% NaOH + autoclave resulted in a greater fiber breakage increasing the cellulose level up to 1.2 times and decreasing 8.5 times the hemicellulose content. This treatment also resulted in a high lignolytic activity for all cultures utilized. T. reesei produced laccase, peroxidase and manganese-peroxidase in all the treatments, having its manganese-peroxidase activity raging from 1.9 to 4.8 times higher than the basidiomycetes.

The study of SOD activity in Jerusalem artichoke tubers in 3 varieties that were different harvesting stage and no report the amount of SOD in different parts of Jerusalem artichoke tuber was carried out. The aims of this study were to evaluate SOD activity in various varieties of Jerusalem artichoke and to evaluate the difference of SOD activity in different parts of Jerusalem artichoke tuber. This experiment used 3 varieties of Jerusalem artichoke including Hel 65, CN 52867 and JA 89 that were different genotypes. These tubers were separated into 3 parts: head, middle and tail for extraction and determination of SOD activity. The results showed that Jerusalem artichoke var.
